He spent his childhood years growing up in Chennai, Tamil Nadu.
He made his Tamil-language film debut as Shiva in the 2006 action film Madrasi.
In 2013, Jagapathi Babu made his first Kannada film appearance as Vijay Kumar in the psychological action thriller film Bachchan.
Alongside N. T. Rama Rao Jr., Rajendra Prasad, and Rakul Preet Singh, he co-starred as Krishnamuurti Kautilya in the 2016 Telugu-language action thriller film Nannaku Prematho. This role won him the SIIMA Award for “Best Actor in a Negative Role (Telugu)”.
His Malayalam-language debut came in the 2016 action thriller film Pulimurugan. He played the role of Daddy Girija and won the SIIMA Award in the “Special Jury Award” category as well as the Asianet Film Award in the “Best Actor in a Negative Role” category.
In 2018, Jagapathi Babu was cast as Basi Reddy, Raghava’s enemy, in the Telugu-language action drama film Aravinda Sametha Veera Raghava. For this role, he received the Filmfare Award in the “Best Supporting Actor – Telugu” category.
He brought to life the character of President Phanindra Bhupathi in the 2018 Telugu-language period action drama film Rangasthalam, starring Ram Charan, Samantha Ruth Prabhu, and Aadhi Pinisetty. For this performance, Jagapathi Babu bagged the SIIMA Award in the “Best Actor in a Negative Role (Telugu)” category.
He has worked with a number of notable directors including S. V. Krishna Reddy, Ram Gopal Varma, Krishna Vamsi, E. V. V. Satyanarayana, Gunasekhar, Chandra Sekhar Yeleti, A. M. Rathnam, K. Raghavendra Rao, Radha Mohan, Mohan Raja, and J. D. Chakravarthy.
In 2019, Jagapathi Babu appeared as Vivek Mittal, a Mumbai-based businessman, in the Telugu-language action drama film Maharshi. It stars Mahesh Babu, Pooja Hegde, and Allari Naresh.
Alongside Chiranjeevi, Nayanthara, Tamannaah, Sudeep, and Vijay Sethupathi, Jagapathi Babu co-starred as Veera Reddy, a chief whose son betrays the resistance, in the 2019 Telugu-language historical action film Sye Raa Narasimha Reddy. The film is inspired by the life of Indian independence activist Uyyalawada Narasimha Reddy.
